L. Dijkhuizen Voorwoord Nog één dag en dan gaat mijn boekje naar de drukker. Het is wel een spannend moment. Alles moet nog een keer bekeken worden om te voorkomen dat er (nog meer) foutjes achter blijven. Het bedenken van de verplichte stellingen en natuurlijk het schrijven van dit voorwoord; de meeste gelezen bladzijde van het hele proefschrift. Juist daarom is deze bladzijde zo belangrijk. Gedurende mijn promotie onderzoek heb ik het geluk gehad om veel mensen te ontmoeten die enthousiast waren over mijn werk en van wie ik veel heb geleerd. De resultaten hiervan zijn o.a. in dit proefschrift te zien. Ten eerste wil ik mijn promoter Lubbert Dijkhuizen bedanken voor de mogelijkheid die hij mij gegeven heeft om bij Microbiële Fysiologie een promotie onderzoek uit te voeren. Lubbert, heel erg bedankt voor jou interesse in mijn werk en ook voor het geduld dat nodig was om mijn manuscripten (weer) te corrigeren. Het heeft jou zeker veel tijd gekost. Bedankt. Zonder de financiële steun van JNICT (Portugal) was ik nooit aan dit promotie onderzoek begonnen. Ik wil daarom de beoordelings comissie van JNICT bedanken voor de goede beoordeling van mijn aanvraag. Mijn speciale collega’s (en vrienden!) Teresa en Hans. Teresa, ik heb altijd genoten van het samen kletsen over allerlei dingen en soms ook over het werk. Jij bent, vooral in het begin, heel belangrijk voor mij geweest toen ik nog niet helemaal aangepast was aan de Nederlandse cultuur. Hans, vanaf het begin heb je mij altijd gesteund in het werk (goede tips) en je bent een goede vriend die altijd klaar stond om mij te helpen. Jij had altijd tijd om te luisteren naar mijn verhaaltjes. Ik wens jou dan ook veel succes in Boston met je nieuwe baan als post-doc. Prefácio Sempre pensei que iria ser mais fácil escrever um prefácio em português, mas falta-me a inspiração e aquela palavra certa para começar. Este prefácio, pode-se melhor traduzir por palavras de agradecimento. Vou tentar por este meio agradecer a todas as pessoas ou instituições que contribuiram de uma ou outra forma para a realização desta tese de doutoramento. A concretização do meu doutoramento só foi possivel devido ao financiamento obtido pela JNICT em Portugal, e ao incentivo dado pelo Prof. N. Van Uden para concorrer a esta bolsa. Mas, um doutoramento não se consegue só com apoio financeiro, o apoio familiar é talvez o mais importante. Ao longo destes 5 anos recebi um apoio muito especial da minha familia não só no que diz respeito ao meu trabalho mas também nos momemtos de adaptação a uma outra cultura. Obrigado. Knowledge of the physiology, biochemistry and genetics of glucose metabolism in actinomycetes is limited. It is well-known that when growth ceases some intermediates from glycolysis and TCA cycle accumulate in the cells, which may serve as precursors for secondary metabolite production. It has been suggested that this represents some kind of overflow metabolism possibly due to lack of strict control mechanisms in these pathways. The aim of this thesis was to characterize enzymes of glucose metabolism and their regulation in actinomycetes. A nocardioform type of actinomycete, the methanol-utilizing bacterium Amycolatopsis methanolica, and the genetically best characterized streptomycete, Streptomyces coelicolor A3(2), were selected for these studies. Characterization of regulatory enzyme steps in glucose metabolism may identify targets for further improvement of industrial strains aiming to enhance availability of precursors from primary metabolism used for production of secondary metabolites. Chapter 1 gives a general introduction of glucose metabolism in bacteria with emphasis on the (limited) knowledge available for actinomycetes; antibiotics formed from intermediates at various positions in the glucose metabolic pathways involved are indicated. The A. methanolica glycolytic enzymes phosphofructokinase, pyruvate kinase and phosphoglycerate mutase are characterized in detail in chapter 2. Also data about glucose and methanol metabolism are presented. The PPi-dependent phosphofructokinase characterized in chapter 2 has been object of further study in chapter 3, describing the cloning and sequencing of the pfp gene and phylogeny of the enzyme. The ATP-dependent phosphofructokinase enzyme from Streptomyces coelicolor A3(2) has been studied in detail in chapter 4. In A. methanolica, a second PFK enzyme (ATP-PFK) inducible by growth on C1 substrates has been characterized (chapter 5). The possible physiological roles for the two PFKs in this organism are discussed. Genetic and biochemical evidence for the presence of a second phosphoglycerate mutase enzyme in A. methanolica is presented in chapter 6. 11
