ANTIBODY SPECIFICITY Mo1 (CD11b) identies the integrin α chain (αM) which is noncovalently associated with CD18.2 It is also the receptor for C3bi.3 Mo1 (CD11b) α chain has a molecular weight of 165 kd and is expressed on monocytes, granulocytes and natural killer cells.4,5,6,7 It is involved in adhesion to endothelial cells and extracellular matrix proteins.2 In neutrophils, Mo1 density increases tenfold as a result of activation by degranulating stimuli.8 Within lymphoid tissues, Mo1 is expressed by histiocytic cells in spleen and lymph node.1 REAGENTS See table above. REAGENT CONTENTS The antibody concentration is 500 µg. The nal concentration of nonantibody reagents when reconstituted is 0.2% gelatin, 0.01 M potassium phosphate, 0.15 M NaCl and 0.1% NaN3. WARNING AND PRECAUTIONS 1. This reagent contains 0.1% sodium azide. Sodium azide under acid conditions yields hydrazoic acid, an extremely toxic compound. Azide compounds should be ushed with running water while being discarded. These precautions are recommended to avoid deposits in metal piping in which explosive conditions can develop. If skin or eye contact occurs, wash excessively with water. 2. Specimens, samples and all material coming in contact with them should be handled as if capable of transmitting infection and disposed of with proper precautions. 3. Never pipet by mouth and avoid contact of samples with skin and mucous membranes. 4. Do not use reagent beyond the expiration date on the vial label. 5. Minimize exposure of reagent to light during storage or incubation. 6. Avoid microbial contamination of reagents or erroneous results may occur. 7. Use Good Laboratory Practices (GLP) when handling this reagent. 8. Harmful if swallowed. 9. After contact with skin, wash immediately with plenty of water. STORAGE AND HANDLING CONDITIONS AND STABILITY RECONSTITUTION PROCEDURE Reconstitute the lyophilized Mo1-FITC reagent by adding 500 µL of distilled water to the vial. This is the stock solution. Centrifuge the stock solution at 20-25°C at 100,000 x g for 10 minutes to optimize staining results. Use this liquid reagent directly from the vial as the stock solution to prepare the reagent working solution. The reagent working solution* is prepared as follows (volume listed is on a per test basis): Add 5 µL stock solution to 195 µL PBS**. *Diluted reagent working solution is good for day of preparation only. **PBS - Phosphate Buffered Saline (pH=7.2). Bring reagent to 20-25°C prior to use. FREEZING PROCEDURE MATERIALS REQUIRED BUT NOT SUPPLIED PBS - Phosphate Buffered Saline (pH=7.2) PN 6603369 PBS containing 2% heat-inactivated fetal or newborn calf serum (FCS). Dilute 2 mL of calf serum to 100 mL with PBS. 1. Dilute the reconstituted stock solution of the COULTER CLONE reagent with PBS containing 2% FCS prior to freezing as follows: Add 5 µL of reconstituted stock solution (1 test*) to 100 µL PBS with 2% FCS**. *These may be frozen in multiple test volume aliquots. **This yields 2x the concentration of the working solution. 2. Prior to use, allow the frozen aliquot to reach 20-25°C. 3. The frozen aliquot, at 2x the nal concentration, must be further diluted to equal the total volume as calculated in the Reconstitution Procedure section. Dilute each aliquot with the appropriate volume of PBS without 2% FCS and mix well. 4. Avoid repeated freeze/thaw cycles. This will denature the antibody protein. 5. Do not store in a self-defrosting freezer. STORAGE AND STABILITY Unreconstituted lyophilized reagent is stable to the expiration date on the vial label when stored at 2-8°C. Do not freeze. Minimize exposure to light. The stock solution of reconstituted lyophilized reagent is stable as follows: • 6 months when stored at 2-8°C or 0 to -20°C when reconstituted using the Reconstitution Procedure. If all of a reconstituted reagent is not to be used within 6 months, follow the Freezing Procedure. • 1 year when stored at -70°C using the Freezing Procedure. EVIDENCE OF DETERIORATION Any change in the physical appearance of this reagent*, or any major variation in values obtained for control samples may indicate deterioration and the reagent should not be used. CONTEÚDO DOS REAGENTES A concentração de anticorpos é de 500 µg. A concentração nal de reagentes não-anticorpos, quando reconstituídos, é gelatina a 0,2%, fosfato de potássio a 0,01 M, NaCl a 0,15 M e NaN3 a 0,1%. ADVERTÊNCIA E PRECAUÇÕES 1. Este reagente contém azida sódica a 0,1%. A azida sódica em condições ácidas produz ácido hidrazóico, um composto extremamente tóxico. Os compostos de azida deverão ser lavados com água corrente, durante a respectiva eliminação. Estas precauções são recomendadas para evitar a acumulação de depósitos na canalização metálica, onde se podem desenvolver condições para explosão. Em caso de contacto com a pele ou olhos, lavar abundantemente com água. 2. Todas as amostras ou colheitas, assim como todo o material que entre em contacto com aquelas, deve ser manipulado como uma substância susceptível de transmitir infecções e eliminado utilizando as precauções adequadas. 3. Nunca coloque a pipeta na boca e evite o contacto das amostras com a pele e membranas mucosas. 4. Não utilize o reagente além da data de validade mencionada no rótulo do frasco. 5. Limitar ao máximo a exposição do reagente à luz durante a sua conservação ou a incubação. 6. Evite contaminação microbiana dos reagentes. Esta pode originar resultados falsos. 7. Respeitar as Boas Práticas de Laboratório (BPL) durante a manipulação deste reagente. 8. Nocivo por ingestão. 9. Após contacto com a pele, lavar imediatamente com água em abundância. CONDIÇÕES DE ARMAZENAMENTO E MANUSEAMENTO E ESTABILIDADE PROCEDIMENTO DE RECONSTITUIÇÃO Reconstitua o reagente Mo1-FITC liolizado, adicionando ao tubo 500 µl de água destilada. Esta é a solução de reserva. Centrifugue a solução de reserva entre 20 e 25°C a 100.000 x g durante 10 minutos para optimizar os resultados de coloração. Utilize este reagente líquido directamente a partir do tubo como solução de reserva, para preparar a solução de trabalho do reagente. A solução de trabalho do reagente* é preparada de acordo com a descrição seguinte (o volume indicado é para um teste): Adicione 5 µl de solução de reserva a 195 µl de PBS**. *A solução de trabalho de reagente diluída só pode ser utilizada no dia da preparação. **PBS - Solução Tampão Salina de Fosfato (pH=7,2). Espere que o reagente atinja uma temperatura entre 20 e 25°C antes da respectiva utilização. PROCEDIMENTO DE CONGELAÇÃO MATERIAIS NECESSÁRIOS MAS NÃO FORNECIDOS PBS - Solução Tampão Salina de Fosfato (pH=7,2) PN 6603369. PBS contendo soro fetal ou de bovino recém-nascido a 2% inactivado pelo calor (FCS). Dilua 2 ml de soro bovino até 100 ml com PBS. 1. Dilua a solução de reserva reconstituída do reagente COULTER CLONE com PBS contendo FCS a 2% antes de congelar, do seguinte modo: Adicione 5 µl da solução de reserva reconstituída (1 teste*) até 100 µl de PBS com FCS a 2%**. *Este volume pode ser congelado em múltiplas alíquotas de volume de teste. **Isto rende 2x a concentração da solução de trabalho. 2. Antes de utilizar, deixe a alíquota atingir uma temperatura de 20 a 25°C. 3. A alíquota congelada, em 2x a concentração nal, tem de ser adicionalmente diluída até igualar o volume total, conforme calculado na secção Procedimento De Reconstituição. Dilua cada alíquota com o volume apropriado de PBS sem FCS a 2% e misture bem. 4. Evite a repetição de ciclos de congelação/ descongelação. Este procedimento irá desnaturar a proteína do anticorpo. 5. Não armazene num frigoríco autodescongelante. ARMAZENAMENTO E ESTABILIDADE O reagente liolizado, não reconstituído, permanece estável até ao m do prazo de validade impresso na etiqueta do respectivo tubo, quando armazenado entre 2 e 8°C. Não congele. Minimize a exposição à luz. A solução de reserva reconstituída do reagente liolizado permanece estável durante: • 6 meses quando armazenada entre 2 e 8°C ou 0 a -20°C quando reconstituída com o Procedimento de Reconstituição. Se não utilizar o reagente reconstituído na totalidade no prazo de seis (6) meses, siga o Procedimento de Congelação. • 1 ano quando armazenada a -70°C, utilizando o Procedimento de Congelação. 4235265–JB 5 of 8 SINAL DE DETERIORAÇÃO Qualquer alteração das características físicas deste reagente*, ou qualquer variação relevante dos valores em relação a amostras de controlo, podem indicar deterioração, pelo que o reagente não deverá ser utilizado.
