Enzimático colorimétrico Determinação quantitativa de colesterol HDL IVD Conservar a 2-8ºC PRINCÍPIO DO MÉTODO Determinação directa do HDLc (colesterol de lipoproteínas de alta densidade) sem necessidade de pré-tratamento ou centrifugação da amostra. O método baseia-se nas propriedades de um detergente que solubiliza só a fracção HDL, de forma que o HDLc liberta-se reagindo com a colesterol esterase, a colesterol oxidase e os cromógenios. As lipoproteínas LDL, VLDL e quilomícrons são inibidas devido à adsorção do detergente nas suas superfícies tornando-as resistentes à enzima. A intensidade da coloração formada é proporcional à da concentração do HDLc presente na amostra testada. SIGNIFICADO CLÍNICO As partículas de HDL são lipoproteínas que transportam o colesterol para as células. O colesterol transportado pelas lipoproteínas de alta densidade é geralmente denominado de “ bom colesterol”, já que niveis elevados estão relacionados com um menor risco cardiovascular. Um nível baixo de colesterol HDL é considerado um dos principais factores de 1,5,6 risco cardiovascular e patologias das artérias coronárias . O diagnóstico clínico deve realizar-se tendo em conta todos os dados clínicos e de laboratorio. CONSERVAÇÃO E ESTABILIDADE Todos os componentes do kit são estáveis, até ao final do prazo de validade indicado no rótulo, quando mantidos nos frascos bem fechados, a 2-8ºC, protegidos da luz e evitando a sua contaminação. Não congelar os reagentes. - R 1 e R 2: Uma vez abertos são estáveis por 8 semanas a 2-8ºC. Não usar reagentes fora de prazo. Indicadores de deterioração dos reagentes: - Presença de partículas e turvação. MATERIAL ADICIONAL - Auto-analisador MINDRAY BS-120 / BS-200E. - Equipamento habitual de laboratorio. AMOSTRAS 1 Soro ou plasma: Não usar anticoagulantes com citrato. Não utilizar amostras hemolizadas. Separar o soro das hemácias o mais rápidamente possível. Estabilidade da amostra: 7 dias a 2-8ºC. VALORES DE REFERENCIA 20/03/14 CONTROLO DE QUALIDADE É conveniente calibrar e analisar juntamente com as amostras os soros controlo e calibradores padrão: SPINTROL H Calibrador, SPINTROL H Normal e Patológico (Ref. 1002011, 1002120 e 1002210). Se os valores determinados estiverem fora do intervalo de tolerância, verificar o equipamento, os reagentes e o calibrador. Cada laboratório deve dispor do seu próprio Controlo de Qualidade e estabelecer correcções caso os controles não cumpram com as tolerâncias. CLINICAL SIGNIFICANCE HDL particles are high-density lipoproteins that transport cholesterol from the body tissues to the liver. Since HDL can remove cholesterol from the arteries and carry it back to the liver for their excretion, HDL is known as “good cholesterol” because high levels are thought to lower the risk of heart disease and coronary artery disease. A low HDL cholesterol levels, is considered a greater heart disease risk1,5,6. Clinical diagnosis should not be made on a single test result; it should integrate clinical and other laboratory data. * Direction Incuba. STORAGE AND STABILITY All the components of the kit are stable until the expiration date on the label when stored tightly closed at 2-8ºC and contaminations are prevented during their use. Do not freeze the reagents. - R 1 and R 2: Once opened is stable 4 weeks at 2-8ºC. Do not use reagents over the expiration date. Signs of reagent deterioration: - Presence of particles and turbidity. SAMPLES Serum or heparinized plasma, free of hemolysis1: Anticoagulants containing citrate should not be use. Removed from the blood clot as soon as possible Stability of the sample: 7 days at 2-8ºC . Blank parameter must be performed in order to get good results in CALIB screen from main menu. The blank calibration is stable until 35 days. After this period the blank parameter must be performed again in order to validate the calibration. QUALITY CONTROL Control sera and calibrators are recommended to monitor the performance of assay procedures: SPINTROL H Calibrator, SPINTROL H Normal and Pathologic (Ref. 1002011, 1002120 and 1002210). If control values are found outside the defined range, check the instrument, reagents and technique for problems. Each laboratory should establish its own Quality Control scheme and corrective actions if controls do not meet the acceptable tolerances.
