Innovative ‘age-defying’ strategy

preoccupation of both women and men. More than being an envelope, the skin is a
complex organ that needs to maintain constantly its physiological functions in order to
ensure homeostasis. Each layer is confronted with certain issues, resolved by specific
solutions. It is thus of prime importance to think of ageing prevention globally, but to
act locally. Soliance has developed an innovative and fully natural strategy that helps
and boosts the biological functions of the skin by acting specifically on each layer of
the skin.
The skin is subject to environmental stimuli and ageing. Both of these events have
harmful biological consequences by disturbing cell homeostasis, and at another level,
whole organ functions. Each layer of the skin is thus subject to biochemical
imbalances. These local issues have to be treated through innovative cosmetic
ingredients that help the skin to recover its normal functions (Fig. 1).
Epidermis
The Stratum corneum can be described as a wall of protection, in which the bricks
represent the corneocytes. The cohesion of the whole is ensured by intracellular
cement which coats the corneocytes, and by complex lipids, the intercorneocyte lipids.
During its growth, Porphyridium cruentum, red microalgae, synthesises a
polysaccharide which creates a protective coating around the cell membrane. Part of
this polysaccharide will dissolve in sea water and increase locally the viscosity. This is a
unique self-protection mechanism. Instead of growing Porphyridium cruentum in sea
water, it is grown in a marine spring water, a unique active water from the coast of
Brittany in France, very rich in various essential minerals which are key cofactors for
skin enzymes. Porphyraline forms a film that protects the skin. By a biomimetic
mechanism, the polysaccharide coats the upper layer of the skin like Porphyridium
cruentum does in nature by secreting the polymer. Thanks to this action, it prevents

the trans-epidermal water loss (TEWL) and helps the skin to keep the water inside.
Porphyraline stimulates the synthesis of skin lipids. This leads to a great improvement
of the intercorneocyte cement. The skin plays more its barrier role and is less subject
to environmental harmful stimuli. The lipid balance is preserved or restored.
Porphyraline helps to repair skin lipid barrier and to maintain skin hydration (In vitro
test)
The experiment is carried out on normal human keratinocytes (NHK). Cells are
inoculated in the epidermisation room. After four days of incubation, a mixture of a
medium of differentiation and the active to test is added to the confluent cells in the
epidermisation room. It is renewed five times every two days for a total treatment
time of 12 days. Lipids are extracted and analysed by chromatography.
Extraction of the lipids and chromatographic analysis: three major lipids of the Stratum
corneum are detected and quantified (average). Porphyraline presents an interesting
stimulating effect on the epidermal lipid synthesis (Fig. 2).
Porphyraline has proved its efficacy in skin moisturising through its barrier role limiting
TEWL, and by reinforcing the Stratum corneum cement. Moreover, this extract is
ECOCERT approved for use in natural cosmetics.
Keratinocytes and ageing
Alteration of lipids due to oxidation is the most well-known example. Recently,
scientific experts have focused on protein damage in the skin. Chronological ageing
and photoageing are characterised by the accumulation of potentially harmful oxidised
proteins that leads to cellular dysfunction and senescence. They are special cellular
targets for reactive oxygen species (ROS).
Each cell contains an intracellular multienzymatic proteolytic system called
proteasome responsible for the degradation of oxidised proteins and protein turnover
(Friguet et al 2000). The proteasome may be found both in cell cytoplasm and nucleus.
The Nobel Prize in Chemistry 2004 was shared between three scientists who made
fundamental discoveries concerning how cells regulate the breakdown of intracellular
proteins with extreme specificity as to target, time and space.
A. Ciechanover, A. Hershko and I. Rose discovered unbiquitin-mediated proteolysis, a
process where an enzyme system tags unwanted proteins with ubiquitin. The targeted
proteins are then transported to the proteasome, where they are degraded by its
three enzymatic proteolytic activities, each cleaving specific peptidic bonds:

LLVY-amc, the chymotrypsin-like enzyme activity.
LLE-na, the post-glutamic-like enzyme activity.
LSTR-amx, the trypsin-like enzyme activity.
Peptides and amino acids are then obtained and may be used again for newly
synthesised proteins.
Oxidised protein build-up is commonly seen as a hallmark of cellular ageing. It is now
well established that an increase of oxidised proteins is associated with a decrease of
proteasome activity in ageing epidermal cells (Carrard et al 2002).
Moreover, harmful effect of UV radiation on skin is related to the generation of ROS
that alter cellular components including proteins. Furthermore, it has been proved that
all the three peptidase activities of the proteasome were impaired after UVA and UVB
irradiation of the cells (Bulteau et al 2002).
Megassane is obtained from lipidic extraction of Phaeodactylum tricornutum. This
microalgae is a marine diatom widely used in aquaculture. Megassane contains a high
content of polyunsaturated fatty acid (PUFAs): C20:5 (Eicosapentaenoic acid) and
C22:6 (Docosa Hexaenoic Acid), both w3 polyunsaturated fatty acid. Soliance has
developed this innovative natural active ingredient that stimulates the proteasome
activity, decreases oxidised protein rate, protects the proteasome activity from UV and
prevents UV-induced damage.
Megassane stimulates proteasome activities and defies chronological ageing (In vitro
tests)
Keratinocytes were treated with 2.5 µg/ml of Phaeodactylum tricornutum extract. The
proteasome activities were assayed in cell lysates using LLVY-amc, LLE-na and LSTRamc as fluorogenic peptide substrate with 20 µg of total proteins. Proteasome
activities were determined as the difference between the total activity of cytosolic
extracts and the remaining activity in the presence of proteasome inhibitor. (Fig. 3)
Phaeodactylum tricornutum extract stimulates the three different proteolytic activities
of the proteasome. It thus helps the epidermis to purify from oxidised proteins.
Oxyblot was performed with protein from keratinocyte lysates after treatment with
2.5 µg/ml of Phaeodactylum (Fig. 4). Treatment with Megassane helps to decrease
oxidised proteins level in NHK with a decrease of 14%.
Megassane restores proteasome activities after UV-irradiation and fights against
photoageing (In vitro tests)
It also has been proved that Megassane protects proteasome proteolytic activities
from UV (Fig. 5).
By restoring proteasome activities after UV exposure, Megassane prevents the
increase of UV-induced oxidised proteins in NHK.
Oxyblot was performed with protein from keratinocyte lysates, after treatment with
2.5 µg/ml of Phaeodactylum extract (cells are previously irradiated with UV or not)
(Fig. 6).
Treatment with Megassane helps to decrease oxidised proteins level in NHK after UV
exposure, with a decrease of 45%.
Megassane maintains skin homeostasis and ensures cell purification. This innovative
active prevents photoageing and may be a new concept of solar protection, in synergy
with filters and anti-oxidants. It may prevent the occurrence of ageing spots where a
constituent (lipofuscine) may contain oxidised proteins.
Dermo-epidermal junction
The dermo-epidermal junction (DEJ) is a mechanical support which links dermis to
epidermis. It is mainly composed of collagen IV and anchoring structures:
hemidesmosomes and anchoring fibrils (composed of collagen VII). Chronological and
extrinsic ageing leads to a decrease of collagen IV and VII synthesis (Craven et al 1997)
and an increase of Matrix Metallo Proteinase 9 (MMP-9) activity (Kähäri, SaarialhoKere, 1997), an enzyme responsible for collagen IV degradation.
The DEJ is different from other basal membranes because it presents specific
anchoring complexes: the hemidesmosomes. These structures solidly fix the
membrane to the epithelium’s collagen fibrils linked to anchoring plaques in the
dermis. They confer an exceptional solidity and organisation. Hemidesmosomes are
specific structures in epithelial basal membranes. Without them, the skin loses its
cohesion, and the dermis and epidermis come apart. They are composed of
intracellular plaques and transmembrane components. The hemidesmosomes play an
essential role in anchoring basal keratinocytes to basal membrane, therefore to the
underlying dermis. They enable the DEJ to ensure its regulating role in water
exchanges, thus guaranteeing the maintaining of skin moisturising.
Ageing is responsible for DEJ weakening (Le Varlet et al 1998), leading to skin firmness
reduction and wrinkle appearance. It is more than necessary to strengthen it day after
day. Le Varlet (1998) studied age-related changes in human DEJ components and
proved that integrin b1 subunit decreases with donor age and that the adhesion of
NHK decreases after the age of 30. As the epidermal anchoring system becomes less
effective with age, this will lead to the occurrence of ageing signs such as wrinkles and
to the production of a lower quality epidermis. Collagen IV is a major component of
the DEJ, both of the Lamina densa, and the anchoring plaque. It is key target of
cosmetic actives (Fig. 7).
Oenothera biennis extract reinforces the DEJ
Oenothera biennis is a plant from Chile brought to Europe during the 17th Century. Its
name, Evening Primrose, comes from the fact that its flowers only bloom during the
night and die the next day. American Indians traditionally used the plant in cataplasms
to soothe wounds or in infusions to treat coughs. Evening Primrose oil is widely used in
cosmetics for its moisturising and restructuring characteristics due to its high content
in polyunsaturated fatty acids. Oenotherol is obtained from the root of Evening
Primrose. It is only developed by Soliance and it reinforces the DEJ through three
complementary actions:

It stimulates the collagen IV synthesis, thus insuring the epidermis basal
membrane stability.
It stimulates the collagen VII synthesis, thus maintaining the skin firmness.
It inhibits the MMP-9 activity responsible for collagen IV degradation.
DEJ integrity is maintained (In vitro test)
The experiment is carried out on NHK. The supernatants containing secreted collagens
are withdrawn after 72 hours of treatment. Collagens IV and VII are dosed by an
adapted ELISA method. (Fig. 8 & Fig. 9).
Oenotherol stimulates Collagen IV and Collagen VII synthesis. It also has been tested
and proved this active inhibits MMP-9 activity, thus protecting components of the
extracellular matrix. Oenotherol is an innovative active ingredient with synergic
properties and a single goal: to maintain the integrity of the DEJ structure. It thus
prevents wrinkle formation. It also prevents premature ageing due to extrinsic factors.
The skin is firmer and looks younger.
Porphyraline increases the number of hemidesmosomes of the DEJ and adhesion of
basal keratinocytes to collagen IV (In vitro tests)
Hemidesmosomes are anchoring structures between dermis and epidermis. They
ensure different biological roles, such as maintaining water flow in the skin.
Porphyraline increases the number of anchoring structures of the DEJ (In vitro test)
Measurement of the number of hemidesmosomes in the DEJ is studied through an
application of Porphyridium cruentum extract (1mg/ml) on NHK and photographic
visualisation. Cells are inoculated in the culture medium. After 2 days of incubation, a
medium of differentiation that contains the active to test is introduced. It is renewed
three times (at D3, D5 and D7) for a total treatment time of 10 days. Observations are
made with optic microscopy that enables the taking of photographs with an
enlargement of x 30,000 (Fig. 10). After application of Porphyraline, the number of
hemidesmosomes is highly increased: +96%. The communication between the dermis
and the epidermis is reinforced.
Porphyraline increases the adhesion of NHK to collagen IV (In vitro test).
In the epidermis, some adhesion molecules are expressed at the surface of basal
keratinocytes. They play an essential role to anchor keratinocytes onto the basal
membrane. The type IV collagen is the main component of the basal membrane,
which, in vitro, binds itself to adhesion molecules of keratinocytes. The study shows
the stimulating effect of on the adhesion of keratinocytes on type IV collagen.
The experiment consists of measuring by protein dosage the adhesion of NHK on a
support covered with collagen IV after one hour of incubation at 37°C. The tested
product is Porphyridium cruentum extract vs control. The adhesion is carried out on
culture plates covered with collagen IV. Cells are left to adhere on the substrate, in the
presence of Porphyridium cruentum extract. The non-adherent cells are then
eliminated by rinsing the holes. The adherent cells are lyzed and quantified by protein
dosage (Kit Sigma, bicinchoninic acid, OD 570 nm) (Fig. 11).
Dermis
The dermis is mainly composed of collagen and elastin. Collagen makes up 70%-80% of
the dry weight of the skin and gives the dermis its mechanical and structural integrity.
During ageing, the synthesis of collagen gradually declines, and the skin becomes
thinner, less stretchable, less resilient, more lax, and prone to wrinkling. In order to
limit the “ageing signs”, Soliance had developed Dakaline, a new active ingredient
obtained from the bark of an African tree, Anogeissus leicarpus. This active helps
collagen synthesis through an innovative mechanism. Vitamin C, known to be the cofactor of collagen synthesis in fibroblast (Dumas et al 1996), is incorporated from the
extracellular matrix (Liang et al 2001). Unfortunately, vitamin C incorporation
decreases with age (Rocquet, Bonte, 2002) (Fig. 12). Anogeissus leicarpus bark extract
stimulates vitamin C incorporation in fibroblasts. This helps to restore and maintain a
normal collagen synthesis level.
Dakaline stimulates vitamin C incorporation in fibroblasts (In vitro test)
Human fibroblasts are treated with Anogeissus leicarpus extract and incubated during
48h. After adding C14 ascorbic acid solution and a 4h-incubation, the cells are lyzed
and radioactivity is measured by scintillation (Fig. 13).
By increasing vitamin C incorporation into fibroblasts, Dakaline helps to stimulate
collagen synthesis. In addition, Anogeissus leicarpus bark extract prevents enzymatic
degradation in the extracellular matrix of the dermis due to MMP-2 and MMP-9. It acts
also as a strong anti-oxidant. Therefore, Dakaline is an innovative active ingredient
which contributes to restore skin homeostasis increases skin firmness.
Moreover, this extract has been developed as a Cosm’Ethic ingredient through a
sustainable development project, by managing the supply chain and local actions in
this way.
Hypodermis
Fat tissues decrease in certain facial areas (Haack, Friedman 2006) due to ageing. This
may result in hollow cheeks and wrinkles. For more than a century, clinicians have
been attempting to supply tissue deficiencies with fat. So, autologous fat
transplantation for soft tissue augmentation has become increasingly popular
(Coleman, 2006). If cosmetic surgery enhances appearance, it is often expensive and
painful. In order to improve women’s wellbeing and beauty, Soliance offers a new and
innovating non-invasive cosmetic solution: Commiphora mukul extract (trade name:
Commipheroline). This new active increases the amount of intracellular lipids by
activating their synthesis and limiting their degradation. Consequently, the adipocyte
volume increases thus reducing depth of wrinkles or toning the bust. Commiphora
mukul is extracted from a shrub growing in the Arabian Peninsula and the arid parts of
Western India. This tree exudates a resin known as Bdellium in the Bible which is
among the oldest fragrant substances. Similar to myrrh, it was traded for centuries by
caravans on the Great Arabian Incense Road. Bdellium was part of the gifts brought by
the Queen of Sheba to King Solomon.
Commiphora mukul extract increases triglyceride level in adipocytes (In vitro test)
Triglyceride level is assayed in vitro on murine pre-adipocytes through NADH dosage (1
mole of triglyceride for 1 mole of NADH) with an enzymatic method (Boehringer
Mannheim). Commipheroline increases the triglyceride level in adipocytes up to 400%
(Fig. 14).
Commiphora mukul extract plumps the breast (In vivo test)
In order to prove in vivo the visible effect of this extract, a clinical study is processed
on breast enlargement. Breasts are a mass of fatty, glandular, and fibrous tissues. As
fat quantity is linked to breast size, a breast enlargement study is a good model to
confirm action mechanism. Commipheroline at 1% in a cream is applied daily on 10
female volunteers (18 to 43 years old females). The breast size is measured by a
dermatologist after 4 and 6 weeks (W4 and W6) of treatment. Individual results at six
weeks show an increase of size from 1 to 4 cm, with a mean value of 2 cm (Fig. 15).
Commipheroline is a new generation of active ingredient. It is able to offer an
alternative to cosmetic surgery.
Conclusion
The skin is naturally sensitive to numerous stimuli and/or damage. Loss of homeostasis
may be due to chronological or premature ageing. Since it is a common wish of men
and women to defy ageing manifestations, cosmetic active ingredients have been
specifically designed to have a role of prevention.
Each biological layer of skin develops its own physiological issues. They all must be
considered as different targets, with specific solutions. However, they also are part of a
whole organ, which is the skin. Innovative age-defying solutions must care about local
problems, whatever may be the cause(s), with a global and holistic point of view.

TESTES CLÍNICOS
1 – Aumento e rigidez dos seios (in vivo) Materiais e Métodos
Foi utilizado Gel de Carbopol com Kigeline à 20%, aplicado uma vez ao dia com massagem
em toda região dos seios e pescoço durante 3 semanas. Foi testado em uma mulher com 26
anos após uma gravidez há mais de 3 anos.
Foram feitos moldes de silicone e fotos antes do tratamento e depois do tratamento (21
dias).
Resultados
Uma modificação na forma e na tonicidade dos seios pode ser avaliada no final do teste. O
seio testado, visivelmente flácido inicialmente, está agora levantado, mais rígido. O mamilo
está mais centralizado e a curva abaixo dos seios diminuída.
O ângulo aberto mostra a diminuição da curva do seio abaixo do seio estando
este mais erguido.
Aumento de 70,52% na elevação dos seios, após 21 dias de uso

Mechanism of action of Commiphora Mukul extract
Commipheroline is extracted from the root of an Indian tree well-known in
Ayurvedic medicine: Commiphora mukul. It presents a plumping effect with two
main applications: wrinkle filling for face care and plumping action specific zones
(lips, breast).
Skin aging leads to alterations of the skin mechanical properties. The adipose tissue
reduction, coupled with the degradation of the extracellular matrix components
make the face skin thinner and thinner. Commipheroline helps to maintain the skin
tone by acting on two enzymes committed in lipogenesis and in lipolysis chain
reactions.
Increase of triglyceride level in adipocyte (in vitro Test)
Triglyceride level in adipocytes increases thanks to G3PDH activation and AMPc
production inhibition. Triglyceride level is assayed 11 days after treatment.
By increasing triglyceride level, Commipheroline helps to fill up wrinkles

Aumento dos seios sem cirurgia
Commipheroline® é um princípio ativo utilizado em cremes que aumenta os seios 2 a 4 cm e atua nas rugas e
flacidez por meio do preenchimento da pele.
Os dados apontam que cada vez mais a mulher procura uma clínica de cirurgia plástica com o objetivo de
aumentar os seios. Entre as jovens adolescentes, a procura aumenta 10% a cada ano. Já as jovens entre 14 e
18 anos representam 15% das operações plásticas. Mas, os especialistas afirmam que a idade ideal para este
tipo de cirurgia é a partir dos 18 anos. Antes disso, as jovens devem tentar outras alternativas, o que até hoje
era impossível em se tratando de aumento de seios, por exemplo.
