FGFR1
Das FGFR1 Protein (fibroblast growth factor receptor
1) gehört zu einer Familie von Rezeptortyrosinkinasen mit zur Zeit vier charakterisierten Mitgliedern
(FGFR-1, -2, -3, -4). Nach bisherigen Untersuchungen
kann eine onkogene Aktivierung der FGFR-Gene
durch Gen-Amplifikation, Translokation oder auch
Punktmutation erfolgen.
Amplifikationen des FGFR1 Gens finden sich in ca.

20 % der Plattenepithelkarzinome der Lunge. Weitere Tumorentitäten die FGFR1-Amplifikation aufweisen können umfassen orale Plattenepithelkarzinome, Plattenepithelkarzinome des Ösophagus und
Mammakarzinome (siehe Tabelle).
Die Wirkung von FGFR-Inhibitoren wie z.B. von Dovitinib (TKI258), einem Inhibitor für FGFR1, FGFR2 und
FGFR3, wird aktuell in ersten klinischen Studien an
Tumoren mit FGFR-Amplifikation untersucht.
