Oxidative stress is a disparity between the rates of free radical production
and elimination through endogenous antioxidant mechanisms such as the
enzymes superoxide dismutase (SOD), glutathione peroxidase (GPx), and
catalase (CAT). This study evaluated the effect of pyrazole compound (5-hydroxy3-methyl-5-trifluoromethyl-4,5-dihydro-1H-1-(2-hydroxybenzoyl) pyrazole) (100,
300 or 1000 µmol/kg, i.p.) on the cogniti ve damage induced by scopolamine and
oxidative stress parameters in rat brain. A step-down inhibitory avoidance
apparatus was used to evaluate the effect of scopolamine (1.4 mg/g i.p.) on
memory function, following biochemical assessments of brain thiobarbituric-acid
reative substances, non-protein thiol groups, SOD and CAT activities, as oxidative
stress biomarkers. Two-away ANOVA did not show a significant effect of
scopolamine or pyrazole treatment on brain thiobarbituric acid reactive
substances, non-protein thiol groups levels, SOD or CAT activities. Statistical
analyses (Kruskal-Wallis H test) showed a significant effect of pyrazole compound
in the recovery from amnesic state caused of scopolamine injection (H = 7,
P<0.05). Post-hoc analysis revealed that 300 µmol/kg pyrazole prior to
scopolamine administration, prolonged step-down latencies compared to the group
that received only scopolamine. The pyrazole compound was effective to prevent
scopolamine-induced amnesia and this effect was not related to a possible
antioxidant activity. This compound could be further evaluated in other models of
dementia.
