Sacha inchi (Plukenetia volubilis L.) is a plant of the Euphorbiaceae family, which grows in the
Amazonian forest. Nowadays it is widely cultivated in the south of Colombia and is becoming a
great object of study due to its protein quality, oil composition and excellent content of omega-3
essential fatty acid. Sacha inchi also presents several kinds of vitamins and minerals in its
composition that are essential for biological system maintaining and they participate as
cofactors in metabolic reactions. This study evaluated the tocopherol composition of Sacha
inchi peanuts and shells. Samples were saponified with a solution of potassium hydroxide KOH
and ethanol, tocopherol fractions were extracted using organic solvent hexano. The isomers of
tocopherols were separated by High Performance Liquid Chromatography fitted with a diode
array detector and C18 reversed-phase column, using an isocratic elution. The solvent flow rate
was maintained at 0.80 mL/min, the column temperature at 25°C and a backpressure of 10 bar.
The mobile phase consisted of methanol:dichloromethane at a ratio of 85:15 (v/v). The UV
detection wavelength was set at 292 nm. Tocopherols were identified by comparison of the
retention times with those of authentic standards from Sigma (USA). Peak areas were
determined using the software Star 6.0 (Varian, USA) by calculated through method by external
standard. Sacha inchi nuts presented delta tocoferol (2.95mg/100g of nut), gama-tocoferol
(4.99mg/100g of nut) and alfa-tocoferol (0.95mg/100g of nut) while Sacha inchi shell presented
0.56, 0.65 and 1.83mg of delta, beta-gama and alfa tocoferol, respectively, per 100g of shell.
