The microorganisms has performed as a promising source of biotechnological
exploitation. Secondary Metabolites produced by microorganisms represents a huge
and inexhaustible source of compounds with therapeutic properties. Therefore, this
work aims apply the versatility of mass spectrometry in the study of the differentiation
of metabolic profiles and dereplication of crude extracts produced by actinomycetes
isolated from mangrove crab shell.
Was cultivated approximately 50 actinomycetes in Erlenmeyer flasks with 25 mL
of the medium BD (potato, dextrose), cultivated at 30ºC, 150 rpm for 10 days. The
cultures where submitted successive liquid-liquid extraction with ethyl acetate (1:1,
v/v). The crude extracts, where analyzed by mass spectrometry, by direct insertion
with flow of 0,150 mL/min, containing 20% of fase A (water/formic acid 0,1 % (1:1))
and 80% of fase B (methanol/formic acid 0,1 % (1:1)). The fingerprints where
acquired in “full scan” mode (ESI-MS) in positive mode and analyzed by Marklynx
program.
The comparative analyze of the chemical profile obtained in the mass spectra
allowed the the agroupment of some extracts who presented similar chemical profile.
The used methodology at this work allowed the identification and characterization of
the surfactin in 6 studied crude extract. Surfactin is lipopeptides with different
biological activities.
