EINLEITUNG TESTKITKOMPONENTEN Die Aviäre Influenza (Geflügelpest) ist eine durch Aviäre Influenzaviren vom Typ A (Familie Orthomyxoviridae) hervorgerufene, anzeigepflichtige Tierseuche, die weltweit Hühner, Puten, Gänse, Enten, wildlebende Wasservögel und andere Vögel infizieren kann. Die klinischen Symptome reichen von milder Verlaufsform (Abfall der Legeleistung und / oder Fertilität) bis hin zu hochpathogener Verlaufsform mit plötzlichen Todesfällen. Die klassische Geflügelpest, ausgelöst durch die Subtypen H5 und H7, gilt dabei als besonders schwere Verlaufsform der Aviären Influenza. Darüber hinaus gilt v. a. H5N1 als Auslöser der Aviären Influenza („Vogelgrippe“) beim Menschen. Bei der klassischen Geflügelpest treten die klinischen Symptome plötzlich und in sehr heftiger Form auf (v. a. respiratorische Symptome, blutiger Nasen- und Rachenausfluss, aber auch grünliche Durchfälle, Zyanosen und Ödeme im Kopfbereich sowie an Kamm und Kehllappen, Verfärbungen an Schenkeln und Füßen bedingt durch Ekchymosen) bis hin zu plötzlichen Todesfällen mit Mortalitätsraten bis zu 100 %. Der FASTest® AIV Ag eignet sich als Screeningtest zum schnellen und einfachen Nachweis aller AIV Typ A-Subtypen in verdächtigen Geflügelbeständen oder bei verdächtigen Wildvögeln und ist vor Ort einsetzbar. Dies ermöglicht dem Tierarzt eine Vor-Ort-Bestätigung seiner klinischen Verdachtsdiagnose und die sofortige Einleitung tierseuchenrechtlich vorgeschriebener Maßnahmen zur Abwehr einer drohenden Pandemie. 1 Testkit FASTest® AIV Ag enthält: – 20 Teststreifen, beschichtet mit monoklonalen Antikörpern – 20 Probenröhrchen mit je 1,0 ml Pufferlösung – 20 Teströhrchen (in Ständern) – 20 Probentupfer – 20 Einmal-Kunststoffpipetten – 1 Gebrauchsinformation HALTBARKEIT UND LAGERUNG Verwendbar bis – siehe Etikett Lagerung 15–25 °C Testkit zum qualitativen Nachweis des Aviären Influenzavirus Typ A-Antigens (Subtypen H1–H15) im Kot oder Kloakenmaterial von Vögeln ANWENDUNG UND ABKÜRZUNGEN GEBRAUCHSINFORMATION In vitro Diagnostikum Die deutsche Gebrauchsinformation ist nach §17c TierSG zugelassen. ABLESEN DES TESTERGEBNISSES 3. INFORMATIONEN ZUM PROBENMATERIAL 5. TESTDURCHFÜHRUNG Den Kot direkt aus der Kloake entnehmen und so schnell wie möglich testen. Auf Grund der i. d. R. inhomogenen oder „nesterartigen“ Verteilung von Antigenen in der Probe vor Probenentnahme muss diese mit Hilfe eines Spatels oder Vortex-Mixers homogen verrührt werden. Beachten Sie, dass das Probenmaterial, ebenso wie alle verwendeten Testkitkomponenten, zum Zeitpunkt der Anwendung Raumtemperatur haben sollte. Endogene und exogene Störsubstanzen einer Probe (z. B. Proteasen, Mucosabestandteile, Blut, aber auch Viskosität, pH-Wert sowie Gräser) können Störeffekte (Matrixeffekte) verursachen, die die Messung des Targets beeinflussen können. Diese können zu gestörtem LF und / oder unspezifischen Reaktionen auf der TL und KL führen. 1. Nehmen Sie die benötigte Anzahl an Teströhrchen. 2. Saugen Sie mit der beigefügten Einmalpipette das PPM komplett und ohne Luftblasen auf. Halten Sie die Pipette senkrecht und tropfen Sie 10 Tropfen in das Teströhrchen (Abb.3). 3. Entnehmen Sie den Teststreifen erst jetzt der Verpackung und stellen Sie diesen senkrecht und in Pfeilrichtung in das Teströhrchen (Abb.4). 4. Bei fehlender Ausbildung der KL nach 30 Minuten muss ein neuer Test angesetzt werden. a. Probentupfer in die Kloake oder in den Kot einführen und so viel Probenmaterial aufnehmen, dass der ganze Tupfer bedeckt ist (Abb.1). b. Den mit Probenmaterial benetzten Tupfer in das mit 1 ml Pufferlösung befüllte Probenröhrchen eintauchen. Mischen Sie die Proben-Puffer-Mischung (PPM) gründlich, bis sie homogen vermischt ist (Abb.2). Abb.2 Das Testergebnis ist nach einer Inkubationszeit von 20–30 Minuten abzulesen. Positive Testresultate können je nach Antigenkonzentration schon früher auftreten. POSITIVES TESTERGEBNIS (Abb.5) Eine schwach bis stark intensiv pink-purpurfarbene TESTlinie und KONTROLLlinie erscheinen. NEGATIVES TESTERGEBNIS (Abb.6) Nur eine pink-purpurfarbene KONTROLLlinie erscheint. Diese Linie zeigt, unabhängig von ihrer Intensität, die korrekte Testdurchführung an. UNGÜLTIGES TESTERGEBNIS Nur eine schwach bis stark intensiv pink-purpurfarbene TESTlinie oder keine der beiden Linien erscheint. VORSICHTSMASSNAHMEN • Beschriften Sie Probenmaterial und zugehöriges Probenund Teströhrchen, damit die exakte Zuordnung gewährleistet ist. • Für jede Probe einen neuen Tupfer, eine neue Pipette sowie neues Proben- und Teströhrchen verwenden. 9. INFORMATIONEN ZUR TESTAUSWERTUNG • Die Interpretation des abgelesenen Testergebnisses sollte im Rahmen der Anamnese, Klinik, Therapie- und Prophylaxemöglichkeiten betrachtet werden. • Die Pufferlösung enthält geringgradige Konzentrationen an toxischem Natriumazid als Konservierungsmittel. Hautkontakt und / oder Ingestion sind unbedingt zu vermeiden! • Jegliche nicht beschriebenen Farb- und Konturabweichungen der TL und KL innerhalb der angegebenen Inkubationszeit bzw. nach mehr als 30 Minuten (z. B. gräuliche, schattenartige Linien) sind als unspezifische Reaktionen und somit als negatives Testergebnis zu werten. • Das Probenmaterial muss als potentiell infektiös angesehen werden und ist mit den verwendeten Testkitkomponenten nach der Testdurchführung fachgemäß zu entsorgen. • Die TL kann sowohl in ihrer Intensität als auch in ihrer Breite variieren und ist daher im Falle eines Erscheinens innerhalb der angegebenen Inkubationszeit als positiv zu interpretieren. 8. TESTPRINZIP Der FASTest® AIV Ag basiert auf einem immunochromatographischen „Sandwich-Prinzip“. Die in der Probe enthaltenen Aviäres Influenzavirus Typ AAntigene (Subtypen H1–H15) reagieren im Bereich des Konjugatkissens mit mobilen, an kolloidale Goldpartikel gebundenen, monoklonalen Anti-AIV-Antikörpern (Anti-AIV-mAKs). Diese Ag-AK-Komplexe durchfließen die Membran („lateral flow“, LF) und werden unter Ausbildung einer m.o.w. intensiv gefärbten pink-purpurfarbenen TESTlinie (TL) an membranfixierte Anti-AIV-mAKs gebunden. Die verwendeten Anti-AIV-mAKs gewährleisten ein hohes Maß an Spezifität zum alleinigen Nachweis von Aviären Influenzavirusantigenen. Die korrekte Testausführung wird durch die Ausbildung einer zweiten, pink-purpurfarbenen KONTROLLlinie (KL) angezeigt. • FASTest® AIV Ag weist alle Subtypen der aviären Influenzaviren Typ A nach, unabhängig von ihrer Pathogenität. • Kreuzreaktionen mit klinisch ähnlich verlaufenden Infektionskrankheiten (z. B. Newcastle Disease Virus (NDV), Infektiöses Bronchitis Virus (IBV), Mycoplasma spp.) sind nicht vorhanden. VORGEHENSWEISE BEI VERDACHT AUF AVIÄRE INFLUENZA 1. Klinisch-epidemiologische Kriterien erfüllt 2. Verdachtsfall: „Aviäre Influenza“ oder „Klassische Geflügelpest“ 3. Testung mittels FASTest® AIV Ag vor Ort a. Negativer FASTest® AIV Ag Wiederholung des Tests bei Fortbestehen des klinischen Verdachts! b. Positiver FASTest® AIV Ag Verdacht auf Geflügelpest erhärtet Meldung (Anzeigepflicht) gemäß tierseuchenrechtlicher Bestimmungen Wichtiger Hinweis Eine Testung vor Ort in verdächtigen Geflügelbeständen oder bei verdächtigen Wildvögeln bedarf grundsätzlich der gesetzlich vorgeschriebenen Bestätigungsuntersuchung in einem mit der Diagnostik der Geflügelpest beauftragten nationalen Referenzlabor. Dies gilt sowohl bei einem negativen als auch bei einem positiven FASTest® AIV Ag. Weitere Informationen erhalten Sie auch beim Friedrich-Loeffler-Institut OIE und Nat. The manufacturer shall not be liable for indirect, special or consequential damages of any kind resulting from the use of this product. Clinical symptoms are mild (decrease of egg production and / or fertility) up to highly pathogenic variants with sudden death cases. The classical avian influenza, caused by the subtypes H5 and H7, is considered to be the severe form of avian influenza. Moreover, H5N1 is considered as elicitor of the avian influenza (“bird flu”) in humans. In the classical avian influenza, the clinical symptoms develop suddenly and heavily (especially respiratory symptoms, bloody nasal and throat discharge as well as greenish diarrhoea, cyanoses and oedemata in the head region and on comb and wattles, changes in colour of thighs and feed due to ecchymosis) up to sudden deaths with mortality rates up to 100 %. FASTest® AIV Ag is suitable as screening test for the fast and simple detection of all AIV type A subtypes in suspicious poultry stocks. It is applicable on-site. This enables the veterinarian an on-site confirmation of the clinical suspicion as well as the immediate initiation of epizootic mandatory measures to prevent a threatening pandemic. 6912 Hörbranz – AUSTRIA www.megacor.com 6. READING OF THE TEST RESULT 3. INFORMATION ON THE SPECIMEN MATERIAL 5. TEST PROCEDURE Take feces directly from the cloaca and test as quick as possible. 1. Take the required amount of test tubes. Due to the normally inhomogeneous or nest-like dissemination of antigens in the sample, the specimen material has to be mixed up homogeneously (spatula, vortex-mixer) before sampling. Keep in mind that the sample material, as well as all used test-kit components, should have reached room temperature at the time of application. Endogeneous and exogeneous interfering substances of the sample (e. g. proteases, mucosa components, blood, but also viscosity, pH-value as well as grass) can cause interferences (matrix effects) that can influence the target measurement. These can lead to an impaired LF and / or unspecific reactions on the TL and CL. 2. Draw the SBM up using the disposable plastic pipette completely and without air bubbles. Hold the pipette vertically and drop 10 drops into the sample tube (fig.3). 20 min Read the test result after 20–30 minutes. Positive test results may be observed earlier, depending on the concentration of antigen in the sample. 3. Remove the dipstick just now from its foil pouch and introduce it vertically and with the arrows pointing downwards into the sample tube (fig.4). POSITIVE TEST RESULT (fig.5) A weak to strong and well-defined pink-purple TEST line and CONTROL line appear. 4. If the CL will not appear after 30 minutes, a new test must be prepared. NEGATIVE TEST RESULT (fig.6) Only a pink-purple CONTROL line appears. This line indicates, irrespective of its intensity, that the test has been performed properly. INVALID TEST RESULT Only a weak to strong and well-defined pink-purple TEST line or no line at all appears. The test should be repeated using a new dipstick. 4. SPECIMEN COLLECTION AND PREPARATION a. Introduce the collection swab into the cloaca or the feces and collect as much sample so that the whole swab is covered (fig.1). b. Immerse the covered swab into the sample tube filled with buffer diluent. INFORMATION FOR THE INTERPRETATION • Label sample material and associated sample tube to ensure a precise assignment. • The interpretation of the test result should always be based on anamnestic and clinical data as well as the therapy and prophylaxis possibilities. • Use a new swab, a new and a new sample and test tube for each sample. • The buffer diluent contains low concentrations of toxic sodium azide as a preservative, therefore avoid skin contact and / or ingestion. • The sample material must be seen as potentially infectious and disposed of accordingly, together with the used test-kit components. 8. TEST PRINCIPLE The FASTest® AIV Ag is based on latest rapid immunochromatographic technique. Positive samples containing Avian Influenzavirus Type A antigens (subtypes H1–H15) react in the conjugate pad area with mobile monoclonal anti-AIV antibodies (anti-AIV mAbs), which are bound to colloidal gold particles. Migrating (“lateral flow”, LF) along the nitrocellulose membrane, these specific antigen-antibody complexes are bound by fixed anti-AIV mAbs producing a more or less intensive pink-purple TEST line (TL). These anti-AIV mAbs guarantee a high level of specificity for the aetiologic detection of AIV. A correct test procedure will be indicated by a second, pinkpurple CONTROL line (CL). • Any non-described colour or contour variation of TL and CL within the indicated incubation time or after more than 30 minutes (e. g. greyish, shadow-like lines) has to be considered as unspecific reaction and therefore as negative test result. • TL can vary both in intensity (from weak to strong pinkpurple) and in width. Therefore, any pink-purple line which appears within the required incubation time has to be interpreted as a positive test result. • FASTest® AIV Ag detects all Avian Influenza Virus Type A subtypes regardless of their pathogenicity. • There are no cross reactions with clinically similar infectious diseases (e. g. Newcastle Disease Virus (NDV), Infectious Bronchitis Virus (IBV), Mycoplasma spp.). PROCEDURE WITH SUSPICION ABOUT AVIAN INFLUENZA 1. Clinical-epidemiological criteria fulfilled 2. Positive FASTest® AIV Ag Suspicion on Avian Influenza substantiated Report according to animal health regulations (duty of notification) Important information An on-site test in suspicious poultry flock or in suspicious wild birds in principle needs a confirmation test prescribed by law in a national reference laboratory that is in charge for the diagnostics of Avian Influenza.
