Receptores 5-HT Serotonin can elicit hyperpolarizing, depolarizing or biphasic responses in its cortical target neurons: Figure 3. Hyperpolarizing and depolarizing effects of 5-HT in layer V pyramidal neurons of medial prefrontal cortex slices from mice. (A) Focal (local) application of 5-HT (100 μM) elicited a hyperpolarizing response which was blocked by the 5-HT1A antagonist WAY 100635 (30 nM), suggesting the expression of 5-HT1A receptors. (B) Focal application of 5HT (100 μM) elicited a depolarizing response which was blocked by the 5HT2A antagonist MDL 11939 (500 nM), suggesting the expression of 5HT2A receptors. (C) Focal application of 5-HT (100 μM) elicited a biphasic response sensitive to the 5-HT2A antagonist MDL 11939 (500 nM). Redrawn from Avesar and Gulledge (2012) with permission from A. Gulledge. From Celada, Puig & Artigas, 2013 5-HT1A receptors are expressed in almost all 5-HT neurons in the mouse rostral brainstem, emphasizing the role of 5-HT1A as an autoreceptor. Neuroquímica do DR 1. The dorsal raphe nucleus (DR) was traditionally believed as a rather homogeneous structure with a global mode of operation. The DR is a highly heterogeneous structure with distinct subregions differentially involved in stress paradigms and affective disorders (Lowry et al. 2008; Hale & Lowry (2011). Frederico Guilherme Graeff (Médico e Pesquisador em Psicofarmacologia), Professor Titular aposentado da Universidade de São Paulo - Campus Ribeirão Preto. The “anxiety” pathway: Early investigations found that interventions resulting in reduced serotonergic neurotransmission increase punished responding, an effect indicative of an anxiogenic role for 5-HT Neuroquímica do DR 96 % The “panic” circuit: Paradoxically, in other animal models of aversive behavior, such as electrical stimulation of the dorsal periaqueductal gray (DPAG), 5-HT has an anti-aversive (i.e. anti-panic) effect. The depression pathway: Finally, it was proposed that a third serotonergic pathway is activated by chronic unconditioned and conditioned stimuli, promoting resilience or tolerance to chronic stress. “the descending reticular system subordinates the lower structures to the control of the cortex.” Luria, A.R. Marcação Neuronal Retrógrada 5HT-2A receptors are expressed in prefrontal neurons projecting to the ventral tegmental area (VTA). As indicated by recent theoretical accounts as well as behavioral studies in animals and humans, 5-HT and DA may have unifying affective, activational and decision functions (Boureau and Dayan, 2011; Cools et al., 2011). Thus, whereas DA primarily serves to promote behavioral activation to seek rewards, 5-HT might be critical for punishment-induced inhibition, promoting withdrawal in the face of aversive predictions (Crockett et al., 2009, 2012; Robinson et al., 2012). Crockett MJ, Clark L, Robbins TW. (2009). Reconciling the role of serotonin in behavioral inhibition and aversion: acute tryptophan depletion abolishes punishmentinduced inhibition in humans. J Neurosci 29:1199311999. A slightly alternative view is that the 5-HT system is critically involved in “waiting to avoid future punishments” and also in “waiting to obtain future rewards” (Miyazaki et al., 2012). Consistent with this, recording studies in presumptive 5-HT DR neurons have indicated that some of them encode behavioral tasks in a systematic manner and encode participation in a behavioral task primarily in terms of its future motivational outcomes (Nakamura et al., 2008; Bromberg-Martin et al., 2010a). Different modes of DA neurons firing: Dopaminergic neurons fire in response to unexpected reward or the conditioned stimulus associated with reward (Schultz, 1998), whereas aversive stimuli or the conditioned stimulus associated with aversive events often inhibit dopamine neuron activity (Matsumoto e Hikosaka 2009; Hong et al. 2011). Schultz, 2001 The dorsal raphe nucleus encodes reward value: “These results suggest that the DRN, probably including serotonin neurons, signals the reward value associated with the current behavior”.
