Glutamato: A glutamina serve como precursora para a síntese de glutamato e aspartato Glutamate is a ubiquitary substance present in most kinds of food and during metabolism: Os sabores básicos: Características ecológicas e importância funcional Têm cinco propriedades básicas do paladar (sabores): amargo, ácido, salgado, doce e “umami” (= glutamato monossódico em particular e aminoácidos em geral). Glutamate is undoubtedly the most prevalent neurotransmitter in the brain. This amino acid is probably being used as a signaling substance in a majority of synapses, alone or along with neuropeptides or other neuroactive compounds that colocalize with glutamate. The excitatory effect of Glu was first recognized in the early 1950s, by Hayashi (1954) and later by Curtis e Watkins (1960), but it took a long time until Glu was generally accepted as a neurotransmitter. Notably, the high concentrations and relatively even distribution of Glu among brain regions were dificult to reconcile with a transmitter role. By the mid-1980s, Glu largely fullfilled the four main criteria for classification as a neurotransmitter: presynaptic localization, release by physiological stimuli, identical action with naturally occuring transmitter, and mechanisms for rapid termination of transmitter action. Introduction of the postembedding immunogold technique to amino acid immunocytochemistry (Somogyi & Hodgson, 1985) made it possible to analyze the distribution of Glu at a quantitative level. This helped distinguish transmitter Glu from other pools of Glu. Later investigations have strengthened a neurotransmitter role for Glu by demonstrating an ATP-dependent selective transport of Glu into purified synaptic vesicles. Hovever, the molecular basis for vesicular accumulation of Glu was long unknown Vesicular glutamate transporters: The discovery of a family of vesicular glutamate transporters (VGLUTs, Bellochino et al., 2000) has opened new possibilities for the identification of glutamatergic neurons. This is the so called Q/R editing site - GluR2(Q) is calcium permeable whilst GluR2(R) is not. Almost all the GluR2 protein expressed in the CNS is in the GluR2(R) form, giving rise to calcium impermeable AMPA receptors. This, along with the interactions with other intracellular proteins, makes GluR2 perhaps the most important AMPA receptor subunit. During normal synaptic transmission, glutamate (Glu) is released from the presynaptic bouton and acts on both AMPA receptors (AMPARs) and NMDA receptors (NMDARs). However, Na+ flows only through the AMPA receptor, but not the NMDA receptor, because Mg2+ blocks the channel of the NMDA receptor. Depolarization of the postsynaptic cell relieves the Mg2+ block of the NMDA receptor channel, allowing Na+ and Ca2+ to flow into the dendritic spine by means of the NMDA receptor. The resultant rise in Ca2+ within the dendritic spine is the critical trigger for LTP.
